产木聚糖酶菌株的筛选及产酶条件优化

         

摘要

通过透明圈法从土壤中筛选到两株产木聚糖酶菌株CS-1和CS-2,CS-2产木聚糖酶的酶活力优于CS-1.CS-2产木聚糖酶最佳发酵培养条件:碳源(甘蔗渣)3.0 g/ml,氮源(NH,CI) 0.5 g/ml,接种量为10%,250 ml三角瓶装液量50 ml,pH=8.0,温度为55 ℃,转速为160 r/min,发酵时间为3d.在此发酵条件下,CS-2产木聚糖酶的酶活力达到7.980 U/ml.%Xylanase producing strains CS-1 and CS-2 were obtained from soil by plate screening method,the xylanase activity of strain CS-2 was superior to the strain CS-1. Meantime, the optimal fermentation condition for producing xylanase with strain CS-2 is as following: the bagasse for carbon source is 3.0 g/mi, the NH4Cl for nitrogen source is 0.5 g/ml,the inoculum dosage is 10%, the liquid volume is 50 ml in 250 ml flask, the pH value is 8.0, the temperature is 55 ℃, the rotary speed is 160 r/min,the fermentation time is 3 days. Under the condition, the xylanase activity was up to 7.980 U/ml.
