秀丽隐杆线虫抗菌药物筛选实验体系的建立

         

摘要

建立了病原菌/药物/生物体三位一体的、多重视频指标的耐药菌敏感的抗菌药筛选评估研究体系,并利用该体系进行耐药病原菌治疗药物筛选.自制用于秀丽线虫培养、观察的多孔板摄影灯箱,用以观察被铜绿假单胞菌感染的秀丽隐杆线虫活动情况.将被感染的线虫置入抗菌药液培养,对秀丽线虫行为模式的视频内容进行记录与统计分析,从线虫死亡率判断药物的疗效.与对照组相比,铜绿假单胞菌感染模型组的秀丽线虫死亡率明显增加,而经抗菌药物(美罗培南)治疗后的秀丽线虫有效降低了死亡率.结论通过对秀丽线虫感染铜绿假单胞菌后的致死率进行评估,建立多重视频指标的耐药菌敏感药物筛选评估研究系统,应用于病原菌感染敏感药物快速简便筛选,及相应的抗菌作用机制研究.%A pathogen/drugs/organism combined drug-resistant bacteria sensitive drug filtrate evaluation research system is established by recording the movements of Caenorhabditis Elegans (C.elegans).A porous plate photographic light box Methodsis constructed for C.elegans culture and observation that C.elegans is cultured in antibiotic solutions and infected by Pseudomonas Aeruginosa (PAl4).Movement of C.elegans is recorded and statistically analyzed.The therapeutic effect is estimated by culture the mortality of C.elegans.Mortality of C.elegans PA14 infected by PA14 increases obviously by contrasting with control group,and then decreases after antibiotic (meropenem).We developed a kind of pathogen/drugs/organism combined equipment for C.elegans-antibiotics filtrate experiment platform.It can be applied in pathogendrug sensitive test and the mechanism research.
