提高细胞融合率的实验方法探讨

         

摘要

Improving the rate of cell fusion is a significant premise of improving the efficiency of preparation of monoclonal antibody and production of new species,and also of improvement of the success rate of new species.This article first puts forward the method of adjusting pH value to improve the cell fusion rate by using PEG 8000 as inducer;its effect precedes PEG 4000.Different cells are used to verify and the best cell density.Using PEG 8000 as inducer and GKN buffer system and toad red blood cells as material,the fusion rate is the highest under the environment of pH8.0 cell fusion.The experiment shows the stabilization and reliability and repeatability are good,and it has the characteristics of user-friendly control and quickness and accuracy.This is a new method of cell fusion,and it can be used in experimental teaching and provide references for scientific research and production.%提出了通过调整pH值的方法,成功将PEG8000作为诱导剂应用于细胞融合过程提高细胞融合率,效果优于PEG4000作细胞融合的诱导剂,使用不同细胞验证并提供了最佳细胞浓度.采用PEG8000诱导,GKN缓冲系统,蟾蜍红细胞为原料,在pH8.0环境下进行细胞融合,融合率最高.实验验证,此方法稳定可靠,重复性好,具有操作简便,快速、准确可重复等优点,为细胞融合实验提供了新方法.可用于细胞融合实验教学,也可为科研、生产提供参考.
