Objective To establish an HPLC method to determine the content of hydroxyacetophenone from different collection time. Methods Supercritical Fluid Extraction was used to extract and HPLC was used to determine the content of hydroxyacetophenone. The HPLC method was performed on a Cromasil C18 column ( 10 μm; 4.6 mm × 250 mm), with methanol-water as mobile phase, the flow rate was lmL/min. the detection wavelength was 275nm and the column temperature was 35 ℃. Results The calibration curve of hydroxyacetophenone was linear in the range of 2～32 tg/mL and the average recovery was 97.47％(RSD=2％).Conclusion This method is accurate, rapid, accurate and reliable with high sensitivity, and can be used for the quality control of Arternisia capillaris.%目的 建立用HPLC测定不同采收期茵陈中有效成分对羟基苯乙酮的含量的方法.方法 采用超声法提取,色谱条件为:色谱柱:Cromasil C18(10 μm,250 mm×4.6 mm),流动相:甲醇-水(35:65);流速:1 mL/min;柱温:35℃;检测波长:275 nm.结果 对羟基苯乙酮在2～32 μg/mL范围内线性关系良好,精密度试验、稳定性试验、重复性试验、加样回收率试验的RSD均小于2%,平均加样回收率为97.47%.结论 该方法准确、快速、灵敏度高,可作为茵陈不同采收期质量控制参考.
