大鼠全脑缺血/再灌注对海马区p63和Bcl-2表达的影响

         

摘要

Objective To observe the effects of global cerebral ischemia/reperfusion on p63 and Bcl-2 expression of hippocampus in rats. Methods 80 male SD rats were randomly divided into two groups: Sham group and global cerebral ischemia/reperfusion (I/R) group, 40 in each group. Rats were executed for cerebral tissues at 6, 12, 24, 48 and 72 h after operation, 8 in each time point. Neuroethology was evaluated. The expressions of p63 and Bcl-2 protein, neuronal density and apoptosis in hippocampus CA1 area were detected. Results The positive ratio of the cells expressing p63 protein in I/R group was significantly higher than that in sham group at the same time point ( P < 0.05 ) , the ratio of 48 h was higher than that of 24 h and 72 h ( P < 0.01). Apoptosis indexes at 24 h, 48 h and 72 h after operation in I/R group were significantly increased compared with the Sham group (P <0.05) , maximum at 48 h. The average gray value of Bcl-2 in 1/ R group was significantly lower than that in Sham group at the same time point (P < 0. 01) , and decreased at 6 h, increased at 12 h, maximum at 24 h ( P < 0. 01). Conclusion The expressions of p63 and Bcl-2 in global cerebral I/R showed a decreasing trend after the first increase, consistent with neuronal apoptosis trend, suggesting that p63 and Bcl-2 might be involved in the regulation of brain neurons.%目的 探讨大鼠全脑缺血/再灌注对海马CA1区p63和Bcl-2表达的影响.方法 80只SD大鼠随机分为假手术组(Sham组)和全脑缺血/再灌注组(I/R组),每组40只.两组分别在手术后6、12、24、48、72 h处死8只大鼠,进行神经行为学评分,检测p63、Bcl-2蛋白表达、神经元密度及凋亡情况.结果 I/R组p63阳性细胞率较Sham组同时相增高(P均<0.05),组内48 h最高(P<0.01);I/R组细胞凋亡指数较Sham组增加(P均<0.05),其中术后48 h最高(P<0.05);I/R组Bcl-2平均灰度值较Sham组低(P均<0.01),组内术后6h开始降低、12 h增加、24 h最高(P<0.01).结论 全脑缺血/再灌注后p63与Bcl-2蛋白表达呈先增加后减少的趋势,与神经元凋亡趋势基本一致,提示p63与Bcl-2可能参与了脑神经元的调控.
