片姜黄产地加工与传统加工在痛经模型中的等效研究

         

摘要

目的 探讨片姜黄产地加工与传统加工在痛经模型中的等效性.方法 将清洁级雌性ICR小鼠分为正常组、模型组(雌激素诱导构造ICR小鼠痛经模型)、阳性药组[痛经宝颗粒2.6g/(kg·d)],鲜切片姜黄高剂量组[3.51g/(kg·d)]、鲜切片姜黄中剂量组[2.34g/(kg·d)],鲜切片姜黄低剂量组[1.17g/(kg·d)],传统片姜黄高剂量组[3.51g/(kg·d)],传统片姜黄中剂量组[2.34g/(kg·d)],传统片姜黄低剂量组[1.17g/(kg·d)].通过记录小鼠扭体反应及前列腺素E2(PGE2),前列腺素F2α(PGF2α),血管加压素(AVP),β-内啡肽(β-EP)五个指标的变化比较两种不同加工方法对治疗痛经的效果.结果 痛经宝颗粒和各给药组扭体次数均显著低于模型组(P<0.01).两种加工方式同剂量给药组比较,产地鲜切加工组微高于传统加工组,但差异无统计学意义(P>0.05).痛经宝颗粒组与两种不同加工方式各浓度剂量片姜黄组的PGE2、PGF2α含量显著低于模型组(P<0.05),两种加工方法随片姜黄给药浓度增高,两者含量呈现减少趋势,但差异无统计学意义,且两种不同加工方式的片姜黄相同浓度梯度剂量差异均无统计学意义(P>0.05).正常小鼠PGF2α/PGE2比值在2倍左右,而模型组小鼠两者比值上升至4倍,两者差异有统计学意义(P<0.01),痛经宝颗粒组与两种不同加工方法不同剂量浓度给药组两者比值均介于2~4,与正常组和模型组比较差异均有统计学意义(P<0.01);痛经宝颗粒组与两种不同加工方式各浓度剂量片姜黄组AVP含量显著低于模型组(P<0.01),β-EP含量显著高于模型组(P<0.05).结论 两种不同加工方法片姜黄均能对痛经起到良好的治疗效果,产地鲜切加工与传统加工等效.%Objective To evaluate the bioequivalence of habitat processed and traditional processed Rhizoma Wenyujin Concisum by the dysmenorrhea model. Methods The estrogen induced dysmenorrhea model of ICR rats were established and the two different processing methods were compared by recording the writhing reaction,prostaglandin E2(PGE2),prostaglandin F2α(PGF2α),vasopressin(AVP),β-endorphin (β-EP)differences for the effect of treatment on dysmenorrhea. Results The number of torsion times of dysmenorrhea granule and each group were significantly lower than that of the model group(P<0.01). Compared with the same dosage group,the fresh cut processing group in producing area was slightly higher than the traditional processing group,but there was no significant difference between the two groups(P>0.05). Dysmenorrhea granule group and the concentration of two kinds of different processing methods and contents of PGE2,PGF2 alpha dose zedoary were significantly lower than that of model group(P<0.05),two methods of processing with turmeric to drug concentration increased,the content was presented to reduce the trend,but there was no significant difference,and two different processing methods of zedoary concentration gradient of the same dose had no significant difference(P>0.05). Normal mice PGF2 alpha/PGE2 ratio was controlled in 2 times,the ratio of mice model both up to 4 times,they were extremely significant difference(P<0.01),while dysmenorrhea granule group and the two different processing methods for different dose concentration ratio between dosage group were between 2 to 4,with the normal group and model group were extremely significant differences(P<0.01). The content of AVP in dysmenorrhea granule group was significantly lower than that in model group(P<0.01)and in turmeric group with different processing methods (P<0.05). Conclusion The two processing methods of Rhizoma Wenyujin Concisum both have good therapeutic effect on dysmenorrhea,and the effect of habitat processed is consistent with that of traditional processed.
