The Naþ aspartate symporter GltPh from Py-nrococcus horikoshii is the only member of the glutamatentransporter family for which crystal structures have beenndetermined. The cation:aspartate coupling stoichiometry isnunknown, thus hampering the elucidation of the ionncoupling mechanism. Here we measure transport ofn22nNaþ and [n14nC]aspartate in proteoliposomes containingnpurified GltPh and demonstrate that three Naþ ions arensymported with aspartate.
