The detailed study of the Pictet-Spengler reaction has progressed from the discovery of improved non-acidic aprotic reaction conditions to probing the mechanistic phenomena involved. We have been able to explore the scope of the condensation including the underlying causes for stereospecificity, the mechanism of epimerization of the C-1 carbon atom, and the optimal conditions for effecting highly stereose-lective reactions. Although these studies are important, their purpose was to provide a gateway through which to access pharmacologically interesting indole alkaloids.
