Inthefaceofemergingmultidrug-resistantmicrobes,reliableanimalmodelsareneededtostudypotentialnewtherapiesininfectedwounds.Tothisend,weimplantedscrew-toptitaniumchamberssubdermallyinfull-thicknesswoundsonbothflanks(n=6perflank)of2Goettingerminipigs.After1wk,chamberswereinoculatedwithStaphylococcusaureus,Pseudomonasaeruginosa,orvehicleonly.Throughoutthestudy,woundfluidwasharvestedforquantitativebacterialculturestomonitorinfection.Animalswerefollowedfor4wk,afterwhichtissuebiopsiesweretakenforhistologicanalysisandquantitativebacterialcounts.Theimplantedtitaniumchamberswerewelltoleratedbythepigsthroughoutthestudy.Afterinoculationofthechambers,woundinfectionwasestablishedandmaintainedfor14d.Despiteinfection,nosystemiceffectswerenoted.Cross-contaminationwasnegligible,comparedwiththevehicle-onlycontrol.Aftertissueingrowth,eachchambercreatesaclosedsystemthatallowsharvestofexudateorapplicationofsubstanceswithoutlossofmaterialfromthechamber.Because12chambersareimplantedineachpig,researchershavetheopportunitytocomparemultipletreatmentoptions(forexample,antibiotics,antimicrobialpeptides,genetherapy)inthesameanimal,withnointerindividualvariation.Weconcludethattheuseoftitaniumchambersinpigsprovidesareliableandreproducibleinvivomodeltoinvestigatewoundhealing,woundinfection,andtreatmentoptions.
