Differential Susceptibility of SD and CD Rats to a Novel Rat Theilovirus

Antibodiestorattheilovirus(RTV)havebeendetectedinratsformanyyearsbecauseoftheirserologiccrossreactivitywithstrainsofTheilermurineencephalomyelitisvirus(TMEV)ofmice.Littleinformationexistsregardingthispathogen,yetitisamongthemostcommonvirusesdetectedinserologicsurveysofratsusedinresearch.Inthestudyreportedhere,anovelisolateofRTV,designatedRTV1,wasculturedfromthefecesofinfectedrats.TheRTV1genomecontained8094nucleotidesandhadapproximately95%identitywithanotherrattheilovirus,NSG910,and73%identitywithTMEVstrains.Inaddition,thegenomesizeofRTV1wassimilartothoseofTMEVstrainsbutlargerthanthatreportedforNSG910.OralinoculationofSprague#8211;Dawley(SD)andCDmalerats(n=10eachgroup)withRTV1revealedthatSDratsweremoresusceptiblethanCDratstoRTV1infection.At14dpostinoculation,100%ofSDratsshedvirusinthefeces,and70%werepositiveforRTVserumantibodies.By56dpostinoculation30%ofSDratscontinuedtohavedetectablevirusinthefeces,and90%hadseroconverted.Incontrast,ininoculatedCDratsRTVwasdetectedonlyinthefecesat14dpostinoculation,atwhichtime40%ofCDratswerefecalpositive.By56dpostinoculationonly20%ofCDratshaddetectableRTVserumantibodies.Ourdataprovideadditionalsequenceinformationregardingarat-specificCardiovirusandindicatethatSDratsaremoresusceptiblethanCDratstoRTV1infection.
