DunkinHartleyguineapigsdevelopspontaneous,age-relatedosteoarthritis(OA)ofthekneeandotherjoints.Histologicchangesareobservedbeginningat3monthsofage.Diseaseseverityincreaseswithage,andat18monthsmoderatetosevereOAisobserved.Astudywasundertakentoassessthemorphologicandbiochemicalchangesof22-month-oldanimals,andtocomparethemwithvaluesin2-month-oldguineapigs.BiochemicalindicescharacteristicofOA,fromtibialcartilage,indicatedanincreaseinproteoglycancontentfrom233±2#59157;g/mg(mean±SEM)at2monthsofageto365±6#59157;g/mgat22months.Collagenconcentrationincartilagedecreasedfrom364±2#59157;g/mgat2monthsto223±3#59157;g/mgat22months.Proteoglycanfragmentsfoundinsynovialfluidmeasured4.6±1g/mlat2monthsandincreasedto37±2#59157;g/mlat22months.Radiographicchangesobservedat22monthsincludedmarginalosteophytesofthetibiaandfemur,sclerosisofthesubchondralboneofthetibialplateau,femoralcondylecysts,andcalcificationofthecollateralligaments.HistologicevaluationrevealedsevereOA,withaMankinscoreof10.7±0.5in22-month-oldanimals.Incontrast,2-month-oldanimalshadnohistologicorradiographicallydetectablelesions.TheresultsofthestudyreportedhereindicatethatthelesionsobservedinthismodelaresimilartothoseofhumanOA.SpontaneousdevelopmentofOAinguineapigsisamenabletothestudyofthepathogenesisofOAandtotheevaluationofpotentialdisease-modifyingagents.
