PlateletabnormalitiesofTesterMoriyama(TM)rats,whichhaveprolongedbleedingtimewithnormalplateletcount,werecharacterizedbycomparisonwiththoseoffawn-hooded(FH)ratswithplateletstoragepooldeficiency(SPD).Morphologically,thedensegranuleswerevirtuallylackinginplateletsfromTMandFHrats.PlateletsfromTMandFHratsaggregatedinresponsetoadenosinediphosphate(ADP),butfailedtohavesecondaryaggregation.Incontrast,plateletaggregationwascompletelyabsentinresponseto1to20#59157;gofcollagen/ml,althoughpartialaggregationwasobservedatthehigherdosageof50#59157;g/ml.NormalamountsofplateletmembraneglycoproteinsIIb/IIIawereexpressedinTMandFHrats,butplateletadenosinetriphosphate(ATP)andADPcontentswerelowerthanthoseinplateletsfromcontrolWistarrats.PlateletATP-to-ADPratioofTMandFHratswassignificantlyhigherthanthatofWistarrats.SerotonincontentinplateletsfromTMandFHratswas20to25%thatofWistarratplatelets.TheseresultssuggestedthatplateletabnormalitiesofTMratsareatypicalcharacteristicofplateletSPDandaresimilartothoseofFHrats,whicharegeneticallydifferentfromTMrats.Therefore,TMratsmayserveasausefulanimalmodelforthestudyofplateletSPD.
