Werecentlydescribedageneticallyengineeredmousemodelthatdevelopsovariangranulosacelltumors(GCTs)thatmimicmanyaspectsoftheadvancedhumandisease,includingdistantdissemination.However,becausetheprimarytumorskilledtheirhostsbeforemetastaseswereabletoform,theuseofthesemicetostudymetastaticdiseaserequiredthedevelopmentofasimple,reliable,andhumanesurgicalprotocolfortheexcisionoflargeGCTsfromdebilitatedmice.Herewedescribeaprotocolinvolvingmultimodalanesthesia,tumorremovalthroughventralmidlineceliotomyandperioperativefluidtherapy,andanalgesiathatledtothepostoperativesurvivalofmorethan90%ofmice,despitetheremovaloftumorsrepresentingasmuchas10%oftheanimal'sbodyweight.IntraabdominalrecurrenceoftheGCTdidnotoccurinsurvivinganimals,butmostdevelopedpulmonaryoradrenalmetastases(orboth)by12wkaftersurgery.WeproposethatthismousemodelofmetastaticGCTwillserveasausefulpreclinicalmodelforthedevelopmentofnoveltreatmentmodalitiesanddiagnostictechniques.Furthermore,ourresultsdelineateanestheticandsurgicalprinciplesfortheremovaloflargeabdominaltumorsfrommicethatwillbeapplicabletoothermodelsofhumancancers.
