Combinationofevaporativedryingandfrozenstorageat#8211;80°ChasbeenusedsuccessfullytopreservehybridB6D2F1mousespermatozoa.Todeterminewhetherthismethodcanbeappliedequallywelltoinbredmice,spermatozoaofC57BL/6JandFVB/NJmicewereevaporativelydriedandstoredfor1moat#8211;80°Cbeforebeingusedforintracytoplasmicsperminjection(ICSI)toproduceliveoffspring.Afterweaning,1maleand1femalemousefromeachlitterwererandomlyselectedat8wkofagefornaturalmatingtoproduceliveoffspring.Resultsshowedthatspermatozoafrombothinbredstrainsthathadbeenevaporativelydriedandsubsequentlystoredat#8211;80°Ccouldbeusedsuccessfullytoderivelive,healthy,andreproductivelysoundoffspringbyICSI.Nosignificantdifferenceswerefoundinembryotransferrate(numberofpupsbornumberofembryostransferred),littersize,weaningrate,bodyweight,numberofpathologiclesions,andamountofcontaminationbypathogensofmiceproducedbyICSIusingevaporativelydriedspermatozoacomparedwithmiceproducedbynaturalmatingorbyICSIusingfresh(thatis,nonpreserved)spermatozoa.ProgenyproducedbymatingmicegeneratedfromICSIusingevaporativelydriedspermatozoawerenormal.Therefore,spermatozoafrominbredmousestrainsC57BL/6JandFVB/NJcanbepreservedsuccessfullyafterevaporativedryingandfrozenstorageat#8211;80°C.
