Becausecrushinjurytoskeletalmuscleisanimportantcauseofmorbidityinnaturaldisasterandbattlefieldsettings,areproducibleandrefinedanimalmodelofmusclecrushinjuryisneeded.Bothopenandclosedsmall-animalmodelsofskeletalmusclecrushinjuryareavailablebutarelimitedbytheirneedforsurgicalisolationofthemuscleorbytheadverseeffectoffibularfracture,respectively.Inthecurrentstudy,wedevelopedandvalidatedanovel,noninvasivemousemodeloflower-extremitymusclecrushinjury.Despitetheclosednatureofourmodel,grossevidenceofmuscledamagewasevidentinallmiceandwasverifiedmicroscopicallythroughhematoxylinandeosinstaining.Theinjuryelicitedbothneutrophilandmacrophageinfiltrationat24and48hafterinjury.TheareapercentageandmeanantigenareaofF4/80-positivemacrophageswerehigherat48hthanat24hafterinjury,andCD68-positivemacrophageareapercentageandmeanantigenareadifferedsignificantlybetweeninjuredanduninjuredmuscle.Inaddition,theincidenceoffibularfracturewasonethirdlowerthanthatreportedforanalternativenoninvasivemodel.Inconclusion,ourmodelisareproduciblemethodformusclecrushinjuryinthemousepelviclimbandisarefinementofpreviousmodelsbecauseofitsdecreasedbonefracturesandreductionofanimalnumbers.
