Epigenetic modulation of microglia modulates the pro-inflammatory cytokines expression in vitro Vaccinia-related kinase 2 is critical for microglia-mediated synapse elimination during neurodevelopment During postnatal neurodevelopment, some of the excessively formed synapses need to be trimmed by microglia in order to com- plete the formation of neural circuits in brain. Lack of synaptic regulation by microglia has been implicated in neurodevelop- mental disorders such as autism, schizophrenia, and intellectual disability. Here we suggest that vaccinia-related kinase 2 (VRK2), which is expressed in microglia, supports synaptic elimination activity of microglia. In VRK2-deficeint mice (VRK2KO), reduced number of presynaptic puncta within microglia was observed. Moreover, the number of presynaptic puncta and synapses were abnormally increased in VRK2KO mice at second postnatal week. The difference did not persist in adulthood. Even though an increase in number of synapses was normalized, adult VRK2KO mice showed behavioral defects in social behavior, contextual fear memory, and spatial memory.
