Ithasbeenshownthat40-60%ofthebacteriafoundindifferenthealthyanddiseasedoralsitesstillremaintobegrowninvitro,phenotypicallycharacterized,andformallynamedasspecies.Thepossibilityexiststhattheseas-yet-uncultivatedbacteriaplayimportantecologicalrolesinoralbacterialcommunitiesandmayparticipateinthepathogenesisofseveraloralinfectiousdiseases.Thereisalsoapotentialfortheseas-yetuncultivatedoralbacteriatotakepartinextra-oralinfections.Foracomprehensivecharacterizationofphysiologicalandpathogenicpropertiesaswellasantimicrobialsusceptibilityofindividualbacterialspecies,strainsneedtobegrowninpureculture.Advancesinculturingtechniqueshaveallowedthecultivationofseveraloralbacterialtaxaonlypreviouslyknownbya16SrRNAgenesequencesignature,andnovelspecieshavebeenproposed.Thereisagrowingneedfordevelopingimprovedmethodstocultivateandcharacterizetheas-yet-uncultivatedportionoftheoralmicrobiomesoastounravelitsroleinhealthanddisease.Keywords:uncultivatedbacteria;oralmicrobiology;molecularbiologymethods;taxonomy(Published:23May2013)Citation:JournalofOralMicrobiology2013,5:21077-http://dx.doi.org/10.3402/jom.v5i0.21077
