MouseoocytesarereversiblyinhibitedfromresumingmeioticmaturationinvitrobycAMPphosphodiesteraseinhibitorssuchas3-isobutyl-1-methylxanthine(IBMX)andcAMPanalogssuchasdibutyrylcAMP(dbcAMP).OocytesculturedinIBMX-containingmediumweretransferredtoandculturedinIBMX-freemediumforvariousperiodsoftimepriortotheirreturntoeitherIBMX-ordbcAMP-containingmedium.Resultsfromtheseexperimentsdefinedaperiodoftimeinwhichoocytesbecamecommittedtoresumingmeiosis.Forskolin,whichelevatedtheintracellularoocytecAMPconcentration,transientlyinhibitedoocytesfromresumingmeiosis.LevelsofcAMPweredeterminedinoocytesincubatedinmediumthatallowsresumptionofmeiosis.ThelevelofoocytecAMPdecreasedsignificantlyduringthetimeinwhichoocytesbecomecommittedtoresumingmeiosis.ThisdecreaseinoocytecAMPwasnotobservedinoocytesinhibitedfromresumingmeiosisbyIBMX.Inaddition,cAMPlevelsweredeterminedinpreovulatoryantralfollicles,cumuluscell-oocytecomplexes,andoocytesduringgonadotropin-inducedresumptionofmeiosisinvivo.AdecreaseinoocytecAMPprecededresumptionofmeiosisasmanifestedbygerminalvesiclebreakdown(GVBD).ThisdecreaseapparentlyoccurredbeforeorduringaperiodoftimeinwhichfollicleandcumuluscellcAMPwereincreasing.AssociatedwithcommitmenttoresumemeiosiswasacharacteristicsetofchangesinoocytephosphoproteinmetabolismthatprecededGVBD.Thesechangesare,todate,someofthefirstreportedbiochemicalchangesthatprecedeGVBD.ResultsfromtheseexperimentsarediscussedintermsofapossiblerolecAMPmayplayinregulationofresumptionofmeiosisinmammals.
