Duringmouseoocytematurationtheregulationoftheactivityofacytoplasmicmaturation-promotingfactor(MPF)wasexamined.ThemouseMPFactivitywasdeterminedbasedonitsabilitytoinducematurationinimmaturestarfishoocytesaftermicroinjectionwiththecytoplasmfrommouseoocytes.MPFappearedinitiallyatgerminalvesiclebreakdown(GVBD),anditsactivityfluctuatedinexactcorrespondencewithmeioticcycles,reachingapeakateachmetaphaseandalmostdisappearingatthetimeofemissionofthefirstpolarbody.CycloheximideaffectedneithertheinitialMPFappearancenorGVBD.Thereafter,however,inthepresenceofcycloheximidethemeioticspindlewasnotformedandMPFdisappeared,althoughthechromosomesremainedcondensed.Afterremovingcycloheximide,MPFreappearedandwasfollowedbythefirstmetaphaseandsubsequentlybypolarbodyemission.Finallythemeioticcycleprogressedtothesecondmetaphase.Thus,fortheappearanceofMPF,thereisacriticalperiodshortlybeforethefirstmetaphase,afterwhichproteinsynthesisisrequired.InthepresenceofeithercytochalasinDorcolcemid,MPFactivityremainedatelevatedlevels.Additionofcycloheximidetosuchcytochalasin-treatedoocytes,inwhichthemeioticcyclewasarrestedatthefirstmetaphase,causedtheMPFlevelstodecreaseandwasfollowedbymovementofchromosomestobothpoleswheretheydecondensedandtwonucleus-likestructureswereformed.Thus,thedisappearanceofMPFmayinitiatethemetaphase-anaphasetransition.Furthermore,detailedcytologicalexaminationrevealedthatchromosomesincytochalasin-treatedoocytesweremonovalentwhilethosetreatedonlywithcycloheximideweredivalent,suggestingthatdissociationofthesynapsisisaprerequisiteforchromosomedecondensationafterthedisappearanceofMPF.Inalltheserespects,MPFseemstobeametaphase-promotingfactorratherthanjustamaturation-promotingfactor.
