Parietalendoderm(PE)contributestotheyolksacandisthefirstmigratorycelltypeinthemammalianembryo.WecanvisualizePEmigrationeminvitro/emusingtheF9teratocarcinomaderivedembryoidbodyoutgrowthsystemand,showherethatPEmigrationisdirectedbythenon-canonicalWntplanarcellpolarity(PCP)pathwayviaRho/ROCK.Basedongolgiapparatuslocalizationandmicrotubuleorientation,68.6%ofcellsincontroloutgrowthsareorientedinthedirectionofmigration.PerturbationofWntsignalingviasFRPtreatmentresultsinalossoforientationcoupledwithanincreaseincellmigration.InhibitionofthePCPpathwayatthelevelofDaam1alsoresultsinalossofcellorientationalongwithanincreaseincellmigration,asseenwithsFRPtreatment.ConstitutivelyactiveDaamcaninhibitthelossoforientationthatoccurswithsFRPtreatment.WepreviouslydemonstratedthatROCKinhibitionleadstoanincreaseincellmigration,andwenowshowthatthesecellsalsolackorientedmigration.CanonicalWntsignalingortheRacarmofthePCPpathwaydoesnotappeartoplayaroleinPEorientedmigration.ThesedatasuggestthePCPpathwayviaRho/ROCKmodulatesmigrationofPE./p/div
