According to the amyloid hypothesis for the pathogenesis of Alzheimer disease, β-amyloid peptide (βA) directly affects neurons, leading to neurodegeneration and tau phosphorylation. In rat hippocampal culture, βA exposure activates tau protein kinase I/glycogen synthase kinase 3β (TPKI/GSK-3β), which phosphorylates tau protein into Alz- heimer disease-like forms, resulting in neuronal death.
