This study evaluated the androgenic potential of 21 Polish cultivars of spring and winter barley. The effects of different culture factors, including pre-treatment, medium and plant growth regulators, were evaluated. The cultivars exhibited different androgenic potential. On average, less than 10 green plants were obtained from 100 anthers (isolated from 3 spikes). Cold pre-treatment of spikes favoured the formation of embryogenic structures (embryos/calluses), most especially in winter cultivars, whereas mannitol pre-treatment enhanced the development of green plants, particularly in spring cultivars. The first embryogenic structures that formed on the induction medium in 4 weeks of culture had the greatest ability to develop green plants, while embryos/calluses collected after 8 weeks of culture were the least efficient. FHG medium supplemented with 1 mg BAP [benzyladenine]/litre and 0.2 mg 2,4-D/litre was optimum for the induction of androgenesis, while the same medium supplemented with 0.4 mg BAP/litre and 3% maltose was optimum for plant development..
