Themechanicalbehaviourofadherentcellsculturedinvitroisknowntobedependentonthemechanicalpropertiesofthesubstrate.Weshowthatthismechanicalbehaviourisalsostronglyaectedbythecells’environment.WefocushereontheimpactoftemperatureandpH.Experimentscarriedoutonindividualcellsinatuneableenvironmentrevealthattheintra-cellularmechanicalbehaviourexhibitslargeandfastchangeswhentheexternalcellenvironmentischanged.Fastpassivemicrorheometrymeasurementsallowfortheprecisecharacterisationofthetransientregimeobservedduringatemperaturedrop.Whenmaintainedatanon-physiologicaltemperature,thecellsreachastabilisedstatedistinctfromthestateobservedinphysiologicalconditions.Theperturbationcanbereversedbutexhibitshystereticbehaviourwhenphysiologicalconditionsarerestored.Thetransientregimeobservedduringtherecoveryprocessisfoundtobedierentfromthetransientregimeobservedwhenleavingphysiologicalconditions.AmodiedgeneralizedStokes-Einsteinequationtakingintoaccountthecellactivitythroughaneectivetemperatureisproposedheretottheexperimentalresults.Excellentagreementbetweenthemodelandthemeasurementsisobtainedfortimelagsfrom103to1sconsideredinthisstudy.
