Apparently unrelated systems often merge and work together to support diverse biological functions. This minireview series aims at unveiling new facets of ESCRTs (endosomal sorting complex required for transport) and calpains (cytosolic cysteine proteases). In the 1990s, yeast genetics revealed a number of genes named Vps (vacuolar protein sorting) whose expression products are involved in transporting integral membrane proteins to vacuoles: the equivalent of mammalian digestive organelles known as lysosomes. Subsequent molecular biological and biochemical studies at the beginning of this century clari?ed that a group of these Vps proteins form four different types of multiprotein complexes known as ESCRTs, and function in recruiting ubiquitylated membrane proteins on endosomal membranes and transporting cargoes to intraluminal vesicles.
