Expression of the Drosophila big brain gene promoter during development

Drosophila big brain (bib) is a neurogenic gene that when mutated causes defects in cell fate determination during Drosophila neurogenesis. The product of bib gene is similar to members of the major intrinsic protein (MIP) family that includes aquaporins, mammalian channels for water as well as other solutes including ions. However the function and expression of the bib gene during development and in adult tissues are little known. In this study, we established transgenic flies bearing the bib-lacZ fusion genes containing the bib promoter region (-2180 to +310 with respect to the transcription initiation site). The bib-lacZ expression of embryos was strongly detected in the nervous system. Among larval tissues, expression of the bib-lacZ gene was detected in the brain and ganglion, imaginal discs, tracheoblast, salivary duct and oenocytes. In adult tissues, high expression of the bib-lacZ fusion gene was detected in the entire brain cortex, antenna, labellum, distal region of legs and oenocytes ofboth sexes, the distal end of testis and the accessory gland cells of males. The bib-lacZ transgenic flies will be useful for further investigation on the regulatory mechanisms of the bib gene.
