We have isolated the genomic DNA for random amplified polymorphic DNA-polymerase chain reaction RAPD-PCR from two geographic lobster species of Korea and the Indian Ocean. The complexity of the banding patterns varied dramatically between the primers and locales from the two locations. We used eight decamer and 20-mer primers to generate a total of 510 fragments in a species of Korean slipper lobster Ibacus ciliatus, and 526 in a species of Indian Ocean deep sea lobster Puerulus sewelli, with DNA fragment sizes ranging from 100 bp to 2,000 bp: 106 specific fragments 20.8% in the Korean lobster species and 136 25.9% in the Indian Ocean species. 242 intra-species-specific fragments, with an average of 30.3 per primer, were observed in the Korean species. 187 fragments, with an average of 23.4 per primer, were identified in the Indian Ocean species. Especially, 132 intra-species-common fragments, with an average of 16.5 per primer, were observed in the Korean species. The OPA-18 decamer primer also generated inter-species-common DNA fragments, with sizes of approximately 150 bp, 200 bp, and 300 bp, respectively, in both the Korean and the Indian Ocean lobster species. The average bandsharing BS value of individuals within the Indian Oceanspecies 0.742 + 0.009 was much higher than in the Korean lobster species 0.710 0.009. The dendrogram obtained with the eight primers indicates two genetic clusters, designated cluster 1 SLIPPER 01 SLIPPER 11, and cluster 2 DEEPSEA 12 DEEPSEA22. Ultimately, the longest genetic distance displaying significant molecular differences was determined to exist between the Korean and the Indian Ocean lobster species genetic distance = 0.612.
