Plant cutin biosynthesis: the involvement of a new acyltransferase

Two important paradigms in plant lipid metabolism relating to plant cuticle genesis are yet to be established. A major component of the cuticle is a lipid polyester called cutin, which in turn is covered by surface waxes. The composition of the cuticular waxes and the insoluble cutin, as well as their physiological characteristics, have been reported previously, but how these compounds are transported out of the cells and through the cell wall is not known and the enzymes involved in cutin biosynthesis have not been characterized. The role of cuticle synthesis in epidermal cell interactions has been described as a new frontier in plant developmental biology. Twenty-seven years ago Rodney Croteau and Pappachan Kolattukudy reported that only leaf epidermis and fruit skin were capable of converting exogeneous hydroxy fatty acid precursors into an interesterified three-dimensional polymer. They postulated the existence of an acyltransferase that transfers the hydroxyacyl moieties from their CoA esters to the growing cutin polymer. To date, no mutants deficient in cutin have been identified for any plant, and no genes involved in cutin biosynthesis are known. Following the hypothesis of this research, we have recently investigated a protein involved in cutin synthesis in epidermis excised from leaves of Agave americana. The cuticle of this plant has been extensively studied in our laboratory.
