Oxygen sensing and adaptive responses to oxygen deprivation play critical roles in many biological processes including cancer, inflammation, and vascular disease. Cells involved in innate and adaptive immunity are key components of the tissue microenvironment in all of these conditions, and their ability to adapt and respond to changes in oxygen levels affects the natural history of pathological processes profoundly. In this issue of the Journal of Leukocyte Biology, Naldini et al. [1] provide further evidence of the involvement of the adaptor protein p66Shc in the response of T cells to a hypoxic microenvironment.
