Given that the Wistar Kyoto (WKY) rat is a putative animal model of depression, the present study examined the effects of acute escitalopram (ESCIT) on the dorsal raphe nucleus (DRN) serotonin (5-HT) neuron and behavior in WKY rats in comparison to Wistar rats. Furthermore, the effect of the repeated ESCIT (5 mg/kg/day s.c. for 14 days) treatment in WKY rats was assessed. Depression-like behavior was evaluated by using forced swim test (FST). Extracellular 5-HT levels were determined from the DRN and the prefrontal cortex (PFC) using microdialysis with HPLC-ECD by means of dual-probe microdialysis. Tryptophan hydroxylase was detected by Western blotting. Neuronal firing activity of DRN 5-HT neurons was assessed using extracellular single-unit in vivo electrophysiology.
