Since its discovery by David Lane in 1979, the tumor suppressor p53 has been consistently a molecule of hotspot. Studies on various aspects of this molecule have led to publication of tens of thousands of papers. The major functions of p53, of course, are the regulation of cell cycle progression and control of apoptosis. Because of its central roles in both cell proliferation and cell death, mutations of this molecule in different sites cause cancers in many types of tissues. In addition, p53 also regulates cell differentiation and is implicated in other human diseases. It regulates the above cellular processes and mediates different human diseases via control of a growing large number of target genes (over one hundred thirty target genes have been identified). In this issue, the first 3 articles reported 4 novel p53 target genes: Bak, c-Maf, Prox-1 and CCL2.
