Heterologous encapsidation of potyviruses in transgenic Nicotiana benthamiana, expressing the coat protein gene of an aphid-transmissible PPV isolate (PPV-AT), is outlined. Several new constructs aimed at minimizing the risks connected with the useof viral genes in transgenic Prunus plants are reported. N. benthamiana plants were transformed by Agrobacterium-mediated gene transfer with PPV genes coding for the P1 protein, the cylindrical inclusion body (C1, helicase), the nuclear inclusion body b(NIb, polymerase) and the coat protein (CP) of a non-aphid transmissible isolate. In addition, modified CP genes leading to nontransferable mRNAs in plants were transformed. To facilitate a rapid screening of transformed tissues, the transferred DNA contains, in addition the viral gene, the gus reporter and the nptll antibiotic resistance gene. Transgenic plants expressing the coat protein gene of an aphid transmissible isolate (PPV-AT) were inoculated with different potyviruses. In all cases, various degrees of PPV coat protein were detected in the newly formed virus particles using polyclonal antibodies directed at PPV.
