Aerial crown-like body formation through nodal segment culture in asparagus

An aerial crown-like body is often formed at the node in asparagus. It is histologically similar to the underground crown that forms at the basal portion of the seedling, with numerous crown buds and vascular bundles with no fixed orientation. Formation of the aerial crown-like body was accelerated by culturing nodal shoot segments of young asparagus cv. Mary Washington 500W seedlings in modified MS medium supplemented with 0.5-2 mg ancymidol/litre and 0.1-0.2 M sucrose. In medium supplemented with2 mg ancymidol/litre and 0.1 M sucrose, aerial crown-like body formation and crown bud induction were very frequent. Aerial crown-like body formation was completely suppressed in the presence of 1-10 mg GA3/litre, and GA3 induced lateral shoot development from the lateral buds. Vigorous rooting was accelerated by transferring the nodal segments with an aerial crown-like body to the basal medium. The plantlets obtained were successfully transplanted to soil.
