The lung has been used as a means for the delivery of locally-acting agents over many centuries for the treatment of lung ailments. Factors which influence the effective delivery of locally-acting small molecules for the treatment of asthma have been reviewed in a recent Advanced Drug Delivery Reviews theme issue . The use of the lung as a portal for the delivery of macromolecular drugs and for the delivery of systemically-acting drugs whether they are small molecules or macromolecules presents some different challenges from those associated with traditional asthma therapy drugs.
