In a double-blind randomised placebo-controlled trial in 366 patients with tuberous sclerosis complex who had frequent partial seizures despite the use of various anti-epileptics, the addition of everolimus reduced seizure frequency. Very few patients became seizure-free. Given its frequent and sometimes severe adverse effects, mainly related to its cytotoxic and immunosuppressive properties, everolimus is only justified for patients with very troublesome frequent partial seizures despite treatment with antiepileptics.
