We described the occurrence and properties of the enzymes engaged in the metabolism of D-amino acid formed from N-acetyl-D-amino acid by the N-acyl-D-amino acid amidohydrolase in Alcaligenes xylosoxydans subsp. xylosoxydans A-6 (Alcaligenes A-6). Oxidase, racemase, and transaminase activities toward D-amino acids were detected, but dehydrogenase activity wasn't. This result suggested that D-amino acid formed from N-acetyl-D-amino acid by N-acyl-D-amino acid amidohydrolase would be further degraded by these enzymes described above. Since Alcaligenes A-6 showed best growth on the medium containing N-acetyl-D-glutamate (D-Glu) among other N-acetyl-D-amino acids (D-aspartate, D-leucine) as a sole carbon and nitrogen source, the metabolism of N-acetyl-D-Glu, in particular, was investigated. D-Glu oxidase activity rapidly increased with the cell-growth re-increasing just after N-acetyl-D-Glu had been consumed in the medium, while high N-acyl-D-Glu amidohydrolase (D-AGase) activity was observed throughout the culture time. It seemed to be considered as the diauxie growth. D-Glu oxidase partially purified, which showed rather high substrate specificity and scarcely act on D-Asp, has been thought to be a membrane-bound enzyme.
