Blockers of KIC1- transporter/channels diminish proliferation of osteoblasticcells

In the preset report, we studied if blockers of the K+/C1- cotransporter, and Cl and 1(± channelsmodify the proliferation of MC3T3-E1 osteoblastic cells. A blocker of the K+/CF cotransporter,DIOA (100 and 5001.1M), diminished the cell proliferation. NPPB (100 and 300 JAM, a blocker ofCI- channel) and quinine (100 and 500 uM, a blocker of K+ channel) also suppressed the prolifer-ation of MC3T3-E1 cells. These blockers of the IC/C1- cotransporter/channels (DIOA, NPPB andquinine) would increase the intracellular CF concentration ([C11). On the other hand, our previ-ous study (Biochem. Biophys. Res. Commun. 361, 1038-1043, 2007) reported that a decrease in[C11, diminished the proliferation of MC3T3-E1 osteoblastic cells. Taken together, the observa-tions indicate that both increases and decreases in [C11, diminish the proliferation of MC3T3-E1osteoblastic cells.
