Comparison of growth of Mycobacterium avium subsp paratuberculosis on three microbiological media [Polish]

Mycobacterium avium subsp. paratuberculosis (MAP) growth is difficult under laboratory conditions. Despite significant progress in research on the isolation of MAP, the only method to reveal viable cells in samples is cultivation on microbiological media. O.I.E recommends three media for isolation and cultivation of MAP: Herrold egg yolk medium (HEYM), Lowenstein-Jensen medium and Middlebrook medium, all of them with the addition of mycobactin J. The aim of the study was to compare the rapidity and intensity of the growth of MAP isolated from contaminated milk samples on the above-mentioned media with and without the addition of antibiotics. The fastest growth of MAP (after 5 weeks of incubations) was obtained on HEYM slops, a little slower MAP growth was on Lowenstein-Jensen slops (6-9 weeks). There was no growth on Middlebrook medium with antibiotics after 16 weeks of incubation. Chemical preparation and decontamination of milk samples allow for the application of microbiological media without antibiotics, which increases the time of MAP growth. [References: 22]
