Properly regulated production of and responses to interleukin 17 (IL-17) are important for the prevention of infection as well as the regulation of susceptibility to autoimmune and inflammatory diseases. Several cytokines (such as IL-6, IL-23 and TGF-beta) and transcription factors (such as RORyt, RORa and STAT3) stimulate the differentiation and population expansion of IL-17-producing cells. IL-17 is produced by conventional CD4~+ T cells (cells of the T_H17 subset of helper T cells), gamma delta T cells and innate lymphoid cells1'2. IL-17 binds to its receptors (IL-17Rs) on many types of stromal cells, which stimulates signaling cascades of the transcription factor NF-kB and mitogen-activated protein (MAP) kinase Jnk and other MAP kinases. Some of these cascades lead to the transcriptional induction of cytokines, whereas others seem to stabilize mRNA.
