We read with interest the recent article (Akcakaya et al., 2007) reporting a quantitative colorimetric method for the measurement of alkaline phosphatase (ALP) activity in eukaryotic cell membranes, especially their results on the relationship between ALP activity of 1G2 hybridomas with respect to the culture conditions. As the serum concentration in the culture media decreased, the authors found that viability of the cells decreased while the specific activity of ALP (measured in OD 405 nm/100 cells) increased. Since a low level or lack of serum leads to nutritional stress, reduced cell viability and apoptosis, the authors suggest that an increase in the ALP activity could be used as an indicator of apoptosis. We show below that this relationship can be described by a simple mathematical function.
