A significant number of eukaryotic genes and regulatory pathways have evolved to ensure that cells replicate and segregate their genomes with high fidelity and at the correct time. Errors in this process result in aneu-ploidy, and the phenotypic consequences are usually profound and often underlie tumori-genesis. The current view is that most spontaneous chromosomal rearrangements result from double-strand breaks (DSBs). A single DSB is lethal if repaired improperly and may result in genetic alterations, which are the leading cause of cancer.
