A less lethal sodium hydroxide test for determining seed coat colour in wheat

Seed coat colour is an important trait in wheat and helps define market class, grade and value of a crop. Current low-technology seed certification tests to check seed coat colour rely on a sodium hydroxide (NaOH) soak which magnifies visual differences between red and white seeds. Unfortunately, NaOH is a strong base and when imbibed by the seed, germination is reduced, limiting further genetic or phenotypic inspection of individual seeds. Here, a modified, less lethal NaOH test is reported. Suggested modifications to the standardised test are a reduced NaOH solution temperature of 21 degrees C and a seed soak time of two minutes, followed by immediate rinsing of seeds under running water. For validation, known red and white wheat kernels were examined visually, by percentage germination and by principle component analysis (PCA) of red, green and blue (RGB) colour values for both standard and modified NaOH soak procedures. The modified test procedure still allows accurate visual, colour-based identification of 'off-type' seeds, and allows germination and DNA extraction so seed certification decisions may be validated by further investigation
