Embryogenic callus formation from protoplasts of Chinese chive (Allium tuberosum Rottl.)

Embryogenic calli were derived from shoot tips of two cultivars of Chinese chive (Allium tuberosum). These calli were used to establish donor cell suspension cultures from which protoplasts were produced. Cell division and colony formation from protoplasts proved to be best in agarose-solidified BDS medium containing glucose. Protoplasts of cv. Wide Green produced microcalli, but those of cv. Super Green Belt did not. Small colonies excised from agarose beads developed into callus on solidified MS medium supplemented with 1 mg/L 2,4-D, and these calli produced somatic embryos on hormone-free MS medium. However, no transferable shoots or plantlets were produced.
