摘要

Whole-cell biotransformation Systems for the conversion of D-fructose to D-mannitol or the reduetion of methyl aceto-acetate to methyl(R)-3-hydroxy butanoate were established in recombinant Escherichia coli strains.Strains construeted contained a recombinant reduction-/oxidation cycle,com-prising NAD-dependent formate dehydrogenase from Mycobacterium vaccae N10(FDH)generating NADH,which was used either by mannitol dehydrogenase from Leuco-nostoc pseudomesenteroid.es ATCC 12291(MDH),or by(R)-speeifie,NADP-preferring alcohol dehydrogenase from Lactobacillus brevis for the respective reduetions.Further-more,for the uptake of D-fructose a glucose facilitator of Zymomonas mobilis(GLF)was additionally expressed in the recombinant D-mannitol producing E.coli strain.
