Production of hairy roots by using Agrobacterium rhizogenes (A4) was achieved using callus tissues of Gloriosa superba as initial explants. The callus tissue was immersed into Agrobacterium rhizogenes suspensions for 30 min infection followed by co-cultivation on MS medium containing Acetosyringone (100 HM) to facilitate hairy root initiation. In addition, the cefotaxime at 500 mg/1 was proved to be efficient for suppression of the growth of A. rhizogenes. Hairy root development was noticed after 3 weeks of culture and they were subsequently transferred into fresh half-strength MS medium for large scale production. Total genomic DNA was isolated from the putatively transgenic hairy roots and control (untransformed tissue) the presence rolA, rolB and rolC genes was confirmed by polymerase chain reaction (PCR) analysis. An efficient method for production of hairy roots in G. superba plants via A, rhizogenes mediated genetic transformation is described and this protocol could be used for production of large scale hairy roots for enhanced level of secondary metabolites extraction for pharmaceutical industry applications in the near future.
