Nucleic acid (I) for producing a polynucleotide (II) encoding a protein (III) for detection and isolation of at least one prey ligand (A) able to bind to a bait polypeptide (B), where (I) encodes a protein comprising a chain of at least two polypeptides (PP), linked (in)directly, where the PP are a detectable marker (PP1) and a sequence (PP2) that binds selectively to a purification support, is new. Independent claims are also included for: (1) a composition containing (I) and a sequence (Ia) that encodes (B); (2) a recombinant vector containing (I); (3) a composition comprising the vector of (2) and a vector that contains (Ia); (4) a recombinant host cell transformed with (I), the vector of (2) or the compositions of (1) or (3); (5) a recombinant host cell containing a nucleic acid that contains, in the same open reading frame, a fusion of (I) and (Ia); (6) a protein comprising a chain of at least 3 PP, i.e. PP1, PP2, and (B); (7) a nucleic acid (IIIa) that encodes (III); (8) a recombinant cloning and/or expression vector containing (IIIa); (9) a recombinant host cell containing (IIIa) or the vector of (8); (10) detecting and isolating at least one (A), produced by a host cell; (11) kits for detecting (A) that bind specifically to (B); (12) detecting (A) present in a sample; and (13) determining a protein interaction, in a host cell, between (B) and native cellular proteins.
