Growth hormone releasing hormone (GHRH) receptor binding was characterized using a unique binding assay using an iodinated GHRH probe. Photo-affinity GHRH probes were made in consideration of photolabeling and characterization of the receptor. In addition, high affinity biotinylated GHRH homologues were made. Dissolution of the GHRH-R / GHRH complex and a gentle wash followed by affinity chromatography extract the specifically bound GHRH resulting in a substantially purified GHRH-R isolate. Electrophoretic treatment of GHRH-R isolates yields GHRH-R of sufficient purity to effect sequencing of the receptor. Cloning of genes encoding polypeptides (proteins or fragments thereof) having GHRH-R activity is performed using a bacterial host, and the cloned genes are expressed in mammalian cell systems.
