PROBLEM TO BE SOLVED: To cultivate a breed by hybridization between distant relative breeds of Oryza sativa, especially to cultivate a remote crossing hybrid rice by discriminating the genotype of S-5 locus by using specific DNA markers.;SOLUTION: The genotype of S-5 locus is discriminated by using DNA markers which are located in the vicinity of S-5 locus which is the hybrid sterility locus of Oriza sativa, and are specified by the base sequences shown by formulas I and II. It is preferable to discriminate the genotype of S-5 locus by carrying out PCR using this set of primers (GTTGGGGTGC AGAAGCGCAT CTTG and ACCGTATGAG GTAGGTTTGA CC) and using DNA extracted from O. sativa to be tested as a template, amplifying a DNA marker located in the vicinity of S-5 locus which is the hybrid sterility locus of O. sativa, followed by determining the genotype of O. sativa to be tested to be S-5n if the amplified DNA marker is cleaved by restriction enzyme XbaI and discriminating the genotype of O. sativa to be S-5i or S-5j if the amplified DNA marker is not cleaved by the enzyme.;COPYRIGHT: (C)2000,JPO
