A short-term anaerobic assay with or without 0.1 g/L chloramphenicol was used to determine the denitrifying enzyme activity (DEA) of two paddy and two upland red soils. The four soils were stored field-moist at ~4℃ ( sample F) , maintained at a soil water content of 40% water holding capacity (sample M) , or saturated with distilled water (sample S) for 110 days. The DEA (μg N2O-N kg-1 h-1) ranged from 2. 15 to 18. 7 in upland soils, and 6. 26 to 327 in paddy soils. For upland soils, submergence increased DEA both with and without the presence of chloramphenicol in the assay ( P <0.01). For paddy soils submergence decreased DEA when chloramphenicol was used, and increased DEA when chloramphenicol was not used (P < 0.05). For the M samples, there were significant differences in the DEA assay with and without choloramphenicol ( P < 0.05 ), but the difference was insignificant for the S samples.
