Class I HLA molecules are nature's proteome scanning chips. Class I HLA sample thousands of intracellular peptides, egress to the cell surface, and then act as peptidepresenting sentries on all cells of the body. Cells of the immune system then (1) survey the peptide cargo presented by these class I HLA sentries, (2) recognize class I HLA presented peptides unique to unhealthy cells, and (3) destroy unhealthy (cancerous and virus-infected) cells. A precise understanding of how class I HLA sentries distinguish diseased cells is positioned to drive immune-based diagnostics, therapies, and vaccines.
