Meningococcal Outer Membrane Vesicles (OMV) are important vaccine candidates against serogroup B meningococci, as these OMVs expose the most important membrane protein (Porin A) that act as primary targets for bactericidal antibodies. The membrane of the OMV is a bilayer composed of different phospholipids. Characterization of these phospholipid constituents is of strategic importance in the OMV production and as part of the validation process of the vaccine product. Due to their amphiphilic nature, chromatographic separation of phospholipids is difficult in terms of separation efficiency, resolution and sensitivity. We present a reversed-phase nanoscale LC-MS technique to determine different phospholipids, based on a system developed in our group and routinely used nowadays for over a decade in peptide and protein analysis.
